I downloaded the application from the official website.
Virustotal shows Trojan.
Is this real?

That would more like to be a false positive.
When you download GlassWire .exe
It is a good practice to also check the file hash

this is the official file downloaded from the site and installed. It shows in the glasswire application after totalvirus scanning. After blocking this file glasswire does not work

Blocking the GlassWire service app will, unsurprisingly, prevent GlassWire from working.

I see the same if I check that file. I would expect Acronis to correct their definitions soon. The reason that TotalVirus has 70+ vendors is so you can make a confidence assessment on the validity of the detection.
